The U. S. Food and Drug Administration (FDA) has warned against the use of an over-the-counter version of the antihistamine (Paliperidone) that was recently introduced as a new treatment for asthma.
The drug, which can cause life-threatening allergic reactions if used in excess, could also be found to be potentially harmful to an unborn baby.
The medication, which is also called Nexium, is prescribed to treat conditions that affect the lining of the womb, called the placenta. It can be found in a variety of products, including some used to treat allergies, heart problems and certain types of cancer.
But a small study published in the journal JAMA Network Open found that the drug, which is sold by a subsidiary of the drug giant AstraZeneca, can also cause a severe allergic reaction in babies that has been seen in infants born to mothers who took the drug during the first two weeks of pregnancy. The symptoms are similar to those of allergic reactions to aspirin or other gastrointestinal medications.
“We have seen an increased incidence of skin rash and anaphylaxis in infants exposed to these products during the first month of life,” said Dr. Steven G. Bleislev, M. D., vice chairman of the Department of Medicine at Mayo Clinic in Rochester, Minn., and professor of pediatrics at Mayo who is lead author on the study. “It’s important to note that this is an extremely rare adverse reaction, so we urge the FDA to immediately stop prescribing Nexium.”
The FDA approved the drug in 2004 for use in the prevention of a type of cancer called squamous cell carcinoma (a type of skin cancer). The drug is now being tested as a treatment for other types of cancer in people who have or were at high risk for developing these types of cancers.
In the study, the researchers looked at a group of people who took Nexium for a year and found that a higher proportion of people exposed to the drug had an allergic reaction than the average person who took one of the drugs on a regular basis.
“Our study shows that over time, patients with the same symptoms who were at high risk of developing a skin cancer and that had used the drug during their first month of life have a higher chance of getting a skin cancer that could be fatal,” said Dr. Bleislev.
In the new study, researchers analyzed data from more than 2,200 people who took Nexium for at least six months and who had either experienced skin rash or anaphylaxis.
The study showed that a group of people who took the drug during their first month of life had a similar rate of skin rash compared to people who had not taken the drug during their first month of life. However, people who had taken the drug during the second and third months of their lives also had a higher likelihood of getting anaphylaxis.
